ARP-11497 is a full-length molecular clone of infectious and replication-competent SIVCPZ provirus generated from overlapping RT-PCR fragments of fecal RNA from a wild chimpanzee without intermediate passage in human or animal host cells (GenBank: DQ374657). The construct consists of approximately 13 kilobase pCR-XL-TOPO®(Invitrogen™) backbone with an approximately 10 kilobase insertion and contains the genes for kanamycin and zeocin resistance.
Note: This plasmid contains the ccdB gene and cannot be propagated in most standard E. coli strains. Plasmids can be propagated in OneShot® ccdB Survival 2 T1R Chemically Competent cells (Invitrogen™) and grown at 37°C. Larger plasmids may benefit from growth at 30°C. This construct may also be grown in other ccdB-resistant competent cells.
Each vial of ARP-11497 contains approximately 5 µg of dried, purified DNA stabilized in DNAstable®Plus (Biomatrica®). Please refer to the appropriate data sheet for lot-specific information.
